Could Long COVID’s Signs of Immune Dysregulation in the Blood Lead to a Diagnostic Test?

STAT—New research has identified proteins present in the blood of people with long COVID that could point the way to a much-needed diagnostic test and possibly to future therapeutic targets. “The dysregulation they observed is a bit complex, as some complement components go up and others go down, but overall the results suggest that complement pathways are abnormal in people with long COVID,” said Gladstone's Nadia Roan, the senior author of a Nature Immunology paper published earlier in January on immune response in long COVID.

Advancing CRISPR Applications: Here, There and Everywhere

BioTechniques—Researchers at Gladstone Institutes have mapped the human immune response using next-generation CRISPR technology known as base editing. Using this tool, the team was able to identify specific nucleotides within T cells that influence immune cells’ response to stimuli, creating maps of DNA sequences and protein sites that are involved in human immune responses. The discoveries this project has facilitated could improve existing immunotherapies and reveal new drug targets for autoimmune diseases and cancer.

‘Striking’ Study Reveals Immune System Anomalies in People with Long COVID

San Francisco Chronicle—At least 10 percent of those who survive COVID continue to face debilitating symptoms months after the acute infection resolves. New research from the laboratory of Gladstone Senior Investigator Nadia Roan shows that individuals suffering Long Covid have a dysregulated immune response to the virus and evidence of T cell exhaustion, which may be due to the virus persisting within their bodies.

Cancer Shredder Utilizes CRISPR, Targets Glioblastoma

Genetic Engineering & Biotechnology News—Scientists at the Gladstone Institutes have published a paper in Cell Reports that describes a CRISPR-based technique for treating primary glioblastoma, a common and aggressive form of brain cancer. The paper, titled “Targeting the non-coding genome and temozolomide signature enables CRISPR-mediated glioma oncolysis,” describes the technique, which the researchers have called “genome shredding.”
